In present study, we discussed the effects of cell microfilament on the activity of collagen type I alpha 1 chain gene (COL1A1) promoter under microgravity simulated by clinostat and/or cytochalasin B as microfilament depolymerizer in the established EGFP-ROS cell line using the method of fluorescence semi-quantitative analysis and the fluorescent stain of microfilament.

但微重力对相关基因调控的影响知之甚少，本研究以构建的基因工程细胞株为对象，以回转器模拟微重力效应，利用增强型绿色荧光蛋白（enhanced green fluorescence protein，EGFP）荧光半定量和细胞微丝荧光染色分析技术，探讨回转模拟微重力条件下，细胞微丝系统对I型胶原α1链基因（collagen type I alpha chain 1，COL1A1）启动子活性的影响。

In this research, the ~ and F2 of the crosses between a natural mutant 慪34?with super-minute grain and 慪38?with super-large grain,慡huhui 881擲huhui 527?with middle grain size were carried out to study the inheritance of the grain shape (grain length, grain width, grain thickness and grain length/width ratio) and 1000-grain weight. The main results are summarized as below:? The F1 grain length, grain width, grain thickness and 1000-grain weight of three crosses were lay between the two parents and tended to Y34, which indicated that those grain traits were all governed by the dominant effect of Y34 and influenced by both female and male parents. The differences of F1 grain length, grain width, grain thickness and 1000-grain weight of positive and negative crosses between Y38 and Y34 indicated the existence of cytogene effects.? The broad heritabilities of major grain traits were calculated.

本研究利用一份水稻极小粒自然突变材料Y34与一份水稻极大粒材料Y38、两份常规籽粒大小材料蜀恢881、蜀恢527的杂交F_1及F_2，对主要粒形性状（粒长、粒宽、粒厚、长宽比）及千粒重进行了遗传研究，根据遗传研究的结果利用微卫星标记结合F_2群分法对控制Y34短粒性状基因进行了分子标记定位，主要结果如下：●各组合F_1粒长、粒宽、粒厚和千粒重介于双亲之间且明显偏向于小值亲本 Y34，这表明粒长、粒宽、粒厚和千粒重均主要受小值亲本显性基因的控制并同时也受大值亲本核基因的影响。Y34与Y38正反交F_1在粒长、粒宽、粒厚、千粒重等性状上存在差异，表明存在细胞质效应。

Positive association were still remained for two locus that MNAT1-rs973063 and PCNA-rs25406 and lung cancer risk even after 1000 times permutation test.During the stratified analysis,we revealed that that several genes in the NER pathway could interact with age,gender,family cancer history and smoking status.

在进行了多重校正后，有两个位点仍显示出关联显著性：MNAT1-rs973063(p=0.02)，PCNA-rs25406(p=0.03)；在随后进行的分层分析中，我们发现NER通路多个基因分别和年龄、性别、肿瘤家族史以及吸烟状况之间存在互作，提示该通路基因位点可能主要是通过和环境交互作用产生效应；我们还进行了单倍型的分析，结果发现PCNA和RPA1两个基因上的单倍型和肺癌易感性相关。

现主要阐述鼻咽癌作为一种多基因遗传性肿瘤的病因发病机制，初步建立鼻咽癌易感基因群的概念和鼻咽癌发生发展过程中易感基因群主导的多阶段性多米诺骨牌效应分子机制假说，为寻找鼻咽癌遗传易感风险因子，筛选鼻咽癌高危人群以及探索鼻咽癌靶向性及个体化的诊断和治疗手段奠定实验与理论基础。

Terminal oxidative inhibitors, such as KCN, Salicylhydroxamic acid , antimycin A and NaN〓, can cut off electron transport chains of cytochrome pathway and alternative pathway, marked decreased BADH protein levels. Oxidative phosphorylation uncoupler 2, 4-dinitrophenol had a similar negative effect on BADH gene expression, and exogenous ATP and ADP increased the BADH levels. These results, in conjunction with the terminal oxidation results, indicated that ATP could be a critical component in BADH gene expression.

2.2 末端氧化抑制剂KCN、SHAM、抗霉素A和NaN〓能打断呼吸链，阻断电子传递，使BADH蛋白的表达量明显降低，说明末端氧化同样参与BADH基因表达的调节：末端氧化影响基因表达的实质就是ATP的影响，线粒体磷酸化解偶联剂DNP明显降低BADH蛋白的含量，而外源的ATP和ADP对BADH基因表达的正效应作用最大。

Results After six passages in BHK-21 cells, cytopathic effect was observed by microscopy. The rescued virus was rejuvenated once in unweaned mice, and then came back to cell passage. We found that the poly tract of rescued virus was shortened when the virus was passaged twice in BHK-21 cells. Although the data of LD50 in mice and TCID50 in BHK-21 cells showed that the virulence and infectivity of genetic engineering viruses were lower than its parental virus, no significant difference was observed between the genetic engineering viruses with the different length poly tract.

我们发现，基因工程毒在BHK-21上连续传代至第6代时，可见明显的致细胞病变效应(cytopathic effect，CPE)；在经过一代乳鼠接种之后再重新适应到BHK-21细胞的过程中，poly区段出现了缩短现象；乳鼠致病性试验和BHK-21细胞TCID50测定结果表明，尽管基因工程毒与母本毒相比毒力偏低，但含有不同长度poly的基因工程毒之间并无显著差异。
