METHODS: The morphological architectures of three HCC cell lines with the identical genetic background and different property of metastasis, including SW620, SW480 (hematogenous and lymphangitic metastasis) and SW480/M, were observed by light microscopy and atomic force microscopy, and the expression of carcinoembryonic antigen, CD44v5, Fascin and proliferating cell nuclear antigen in those cell lines were detected by immunohistochemical method.

Primary culture of bovine corneal endothelial cells and whole rabbit corneas were used as the experimental materials. We first treated the cultured bovine corneal endothelial cells with PTPs inhibitor, sodium orthovanadate, with a variety of concentrations (25,50,100μM) for various durations (8,24 hrs). The effects of PTP inhibition on cellular distribution of cell-cell junctional proteins, such as N-cadherin, alpha-catenin and p120, were evaluated by immunohistochemical staining with fluorescein microscopy and confocal microscopy. Immunohistochemical staining with Ki67 Ab, a marker of cell proliferation, was also used to detect cells entering cell cycle.

The microstructure evolution and composition distribution of the 7A55 cast and homogenized alloy were studied by optical microscopy, scanning electron microcopy, energy dispersive spectrometer, transmission electron microscopy and X-ray diffractometry. The homogenizing and overburnt temperatures of the alloy were determined. Meanwhile, the diffusion kinetics of the alloy elements during homogenizing treatment was investigated and the homogenizing kinetic equation was deduced according to Fick's First Law of diffusion. The diffusion of alloy elements, precipitation of Al3Zr particles and the conglomeration and globe of the insoluble phases were explained by the equation during homogenization.
