观察

Rat in Group B was set an air bladder which was empty and not perfused water. Water of 0.6 ml, 0.8 ml, 1.0 ml, 1.2 ml, 1.4 ml and 1.5 ml was perfused respectively into the air bladders which were set in the colon of rats in group C, D, E, F, G and H. Then the changes of ethology of the rats were recorded in interval of an hour.

应用疼痛行为学的观察方法，观察1h各组动物的行为学改变，并采用免疫荧光化学的方法，观察大鼠DCN中神经元被激活的标志c-fos及胶质细胞的胶质原纤维酸性蛋白(glial fibrillary acidic protein， GFAP)和小胶质细胞特异性补体OX42的表达情况，空白对照组是在疼痛高峰点(1.2 ml)处死取材。

MAIN OUTCOME MEASURES: Cellular proliferation at the 1st, 2nd, 3rd days of cell culture; Alkaline phosphatase activity at the 6th day of cell culture.

主要观察指标：细胞培养第1，2，3天观察细胞增殖情况；第6天观察细胞内碱性磷酸酶活性。

MTT assay FAK signaling pathway inhibitor genistein on human corneal epithelial cell cytotoxicity;RT-PCR detection of human corneal epithelial cells adhesion to fungus at different times,extracellular matrix protein including laminin,fibronectin,FN glass,Ⅳcollagen,transmembrane protein integrinαⅤ,integrinβ1(ITGβ1),as well as the FAK signaling pathway FAK1,FAK2 and Paxillin gene expression;Western blot detection of the signal transduction pathway adhesion-associated protein ITGβ1,FAK and PAX expression and the inhibition of genistein. Immunocytochemical method was used to observe the LN,FN and FAK expression in human corneal epithelial cells during interaction with the fungues;Laser scanning confocal microscope had a cell positioning on FN,FAK and PAX,observed the changing of the human corneal epithelial cytoskeleton after stimulated by fungues;Quantitatived by flow cytometry to detect of human corneal epithelial cells with PAX at ITGβ1 fungal expression after adhesion;Optical microscopy quantitied the fungues and human corneal epithelial cell adhesion and recorded to determination the integral optical density afrer adhesion;Scanning and transmitted electron microscope observed the changing of cell ultrastructure after fungues and human corneal epithelial cell adhesion.

第一部分真菌激活人角膜上皮细胞FAK信号转导通路的体外实验研究将三种常见致病真菌(白色念珠菌、烟曲霉菌和茄病镰刀菌)分别与人角膜上皮细胞共孵育，MTT法检测FAK信号通路抑制剂染料木黄酮的对人角膜上皮细胞的细胞毒性作用；RT-PCR检测真菌黏附人角膜上皮细胞后不同时间细胞外基质层连蛋白、纤连蛋白、玻连蛋白、Ⅳ型胶原、跨膜蛋白整合素αV、整合素β1(ITGβ1)，以及FAK信号通路中FAK1、FAK2和桩蛋白基因的表达情况；Western blot的方法检测黏附信号转导途径相关蛋白ITGβ1、FAK和PAX的表达，以及染料木黄酮对真菌刺激人角膜上皮细胞FAK信息通路活化的抑制作用；免疫细胞化学方法观察人角膜上皮细胞与真菌相互作用过程中LN、FN和FAK的表达；激光共聚焦显微镜对FN、FAK和PAX进行了细胞定位，并观察真菌刺激后人角膜上皮细胞骨架的变化；流式细胞仪定量检测人角膜上皮细胞ITGβ1与PAX在真菌黏附后表达的改变；光学显微镜观察真菌与人角膜上皮细胞黏附数量，记录并测定了黏附后积分光密度值OD扫描及投射电镜观察了真菌与人角膜上皮细胞黏附后，细胞超微结构的改变。

Histologically,the activity of cell division in scutellar epithelium and callus cluster derived from it was higher in cultivar Aikoku than Moritawase.Moreover different types of differentiation in the epithelial callus were observed,the rootlike structures and globular cell clusters with epidermislike structure were found in the calli of cultivar Moritawase and Aikoku respectively.

组织学的观察表明，Aikoku的种胚盾片上皮组织以及由上皮细胞起源的愈伤组织的细胞分裂活性高于Moritawase；而且在上述2个品种中还观察到不同的细胞分化类型，在Aikoku的愈伤组织中观察到了带有表皮状结构的圆形细胞块，而在Moritawase的愈伤组织中却观察到了根状结构。

Result］all of enzyme tests were normal.the author could observe the dissolved degeneration of part of sacrospinal muscle fibers,muscle fiber hypertrophy,and a small quantity of inflammatory cell infiltration with a light microscope.focal solution of muscle fiber,the aggregation of mitochondria around the nucleus,the increase of lipid droplet and lysosome in cyte,and the proliferation and differentiation of muscle satellite cell could be observed with an electron microscope.

术中切取竖脊肌标本用于组织病理学观察和透射电镜观察。［结果］各项酶学检验无异常；组织学光镜下观察到竖脊肌纤维部分溶解变性，肌纤维肥大，少量炎性细胞浸润；电镜下观察到肌纤维灶状溶解，核周线粒体聚集，胞内脂滴、溶酶体增多，肌卫星细胞增殖分化。

术中切取竖脊肌标本用于组织病理学观察和透射电镜观察［结果］各项酶学检验无异常；组织学光镜下观察到竖脊肌纤维部分溶解变性，肌纤维肥大，少量炎性细胞浸润；电镜下观察到肌纤维灯状溶解，核周线粒体聚集，胞内脂滴、溶酶体增多，肌卫星细胞增殖分化。

Methods 16 epithelials of lens anterior capsules of diabetic senile cataract were divided into two groups,15 were stained with HE and examined with light microscopy.1 was examined with transmissional electron microscope. The same examination in 16 epithelials of lens anterior capsule in senile cataract was carried out.

收集16例糖尿病患者的老年性白内障眼的晶状体前囊膜上皮分为两组，15例进行HE染色，光学显微镜观察，1例制片电镜观察；对照组取16例老年性白内障眼的晶状体前囊膜上皮分为3组，15例进行HE染色，光学显微镜观察，1例制片电镜观察。

Methods: Integrated and disrepaired skins in the rats were smeared with Salicylic AcidPhenol Liniment by the maximum tolerated dose only. The toxic reaction of the animals were observed and recorded continuously from the beginning of using the drug to 7 days, and evaluated by using the general standard of the reaction of skin stimulation in pharmacology. Animals were executed by taking off acanthi and blood was collected to check up the periphery blood routine and serum biochemistry at the end of the observation of the rats. And after the dissection, the pathological change of main visceras, such as heart, brain, liver and kidney were observed with naked eye and pathological slices. The data of this experiment was used to calculate the value of LD50 by using Bliss rule.

观察大鼠完整皮肤及破损皮肤一次性涂抹最大耐受量的小儿水杨酸苯酚搽剂所产生的毒性反应情况，从给药0时起至7 d连续观察记录动物的急性毒性和45 d连续的长期毒性反应，以皮肤刺激反应标准进行评分作毒理等级评价；对存活动物在观察结束时进行高位脱椎处死采血检测外周血常规和血清生化，解剖后裸眼和病理切片观察心、脑、肝、肾等主要脏器的病理变化。
