The colony forming and the cell cycle were analyzed with flow cytometry, and the mRNA expression of bcl-2, c-myc gene were determined with RT-PCR method. Results Resveratrol could markedly inhibit proliferation of human gastric cancer cells compared with that of the blank control group. The cell cycle was blocked to G0/G1. The mRNA level of bcl-2 and c-myc gene were decreased compared with that of the blank control group.

结果 白藜芦醇可抑制胃癌细胞增殖和集落形成能力，低、高剂量组的抑制率分别为6.9%，17.6%和27.98%，63.39%，差异均有统计学意义(P.05)；白藜芦醇能使胃癌细胞周期阻滞于G0/G1期，明显下调胃癌细胞bcl-2、c-myc基因mRNA表达水平(P.05)；用药24 h后，白藜芦醇高剂量组bcl-2和c-myc基因相对表达量分别为(0.54±0.04)，(0.58±0.05)，与空白对照组(0.91±0.05)，(0.85±0.03)比较，差异均有统计学意义(P.05)。

The expression level of metabolitic enzyme (CYP1A1, CYP1B1) and its correlation with cell differentiation /apoptosis were studied as well. The influence of resveratrol in cell cycle was determined with FCM. Results 1 Resveratrol not only suppresses the growth of medulloblastoma cells in a dose and time related fashion but also induces cell differentiation and apoptosis. Resveratrol promotes differention of UW228-1,-2 cells to forward glia, UW228-3 and Med-3 cells to neuron. A treatment of resveratrol in the concentration of 100μM for 48hrs comit most of cells die of apoptosis. 2 Among the four cell lines Fas and Caspase-3 were constantly expessed, whereas FasL was absent irrespective to the drug treatment. 3 ICC, RT-PCR, Western-blot hybridization and EROD enzymatic activity assay demonstrated that expression of CYP1A1 is different after treatment with resveratrol in four cell lines, up-regulated in three UW228 cell lines in a dose dependent manner but down-regulated in Med-3 cells. Suppressive effect of resveratrol on CYP1B1 expression is the same among four medulloblatoma cell lines. 4 Cell cycle distribution of UW228-3 was greatly changed after treatment.

结果：1、白藜芦醇以时间和剂量依赖性方式抑制髓母细胞瘤细胞生长，同时促进细胞的分化和凋亡：白藜芦醇促使UW228-1、-2靶细胞向神经胶质细胞方向成熟分化，UW228-3、Med-3细胞向神经元细胞方向成熟分化，以100μM作用48小时效果显著；2、四个细胞系均有Fas和Caspase-3表达，但无Fas-L基因表达和蛋白活性产生，因此难以形成Fas相关性死亡通路；3、白藜芦醇处理前后CYP1A1基因的表达在各细胞系略有不同：UW228-1，-2，-3三个细胞系白藜芦醇以剂量相关性方式上调CYP1A1的表达，而Med-3细胞系，白藜芦醇却抑制CYP1A1的表达；四个细胞系在白藜芦醇处理后CYP1B1的表达均受到抑制；4、UW228-3细胞系经白藜芦醇作用后细胞周期有较大改变，表现在G0/G1期在整个细胞周期中所占的比例增加，而其它时期则相应减少。

For the enzyme extraction, with the ratio of enzyme/giant knotweed 1:500 at 50℃ for 90 min, the yield of resveratrol is two times more than that extracted by ethanol. For the microwave extraction with the ethanol content of 80%, the liquid ratio of 1:20, extracting by microwave for 60s at 510W, the yield of resveratrol is three times more than extracted by ethanol with enzymation 30 min by microwave for 10 s at 510 W.

酶法辅助提取白藜芦醇的适宜提取条件为：酶解温度为50℃、pH值为5.0、纤维素酶与虎杖粗粉质量配比为1：500、酶解时间为90 min，比传统乙醇提取法提取收率提高2倍；微波辅助醇提白藜芦醇的适宜提取条件为：以体积分数为80%的乙醇溶液为提取剂，料液质量比为1：20，在510 W功率下微波1.0 min，比传统乙醇提取法提取收率提高近3倍；利用酶微波结合提取白藜芦醇，酶解30 min， 510 W功率下微波10 s，提取收率提高3倍，缩短了提取时间，降低了生产成本。

Results: Resveratrol significantly ihibited growth and proliferation of SW480 cells in time and dose dependent manner.Resveratrol perturbed cell cycle,and arrested SW480 cells in S-phase accompanied with reducing G2-phase cells.The ultrastructural change of the SW480 cells treated with resveratrol were as following:the number of microvilli on the surface ofplasm membrance decreased and the nucleolar margination was frequent in nucleus.

结果：白藜芦醇能抑制SW480细胞的生长增殖，并呈时间浓度依赖性；白藜芦醇能影响SW480细胞的细胞周期分布，使细胞周期阻滞于S期，G1期细胞减少；经白藜芦醇处理的SW480细胞可出现细胞超微结构的改变：可见染色体边聚，细胞表面突起减少，胞质内空泡化等凋亡形态的变化。

The results indicated that resveratrol content in different producing areas varied during the fermentation and it increased gradually along with the fermentation and the addition of pectase and it reached the highest level up to chief fermentation, then it decreased and maintained a certain level till malolactic fermentation.

To establish a MPLC method for the content determination of resveratrol in Polygonum cuspidatum Sieb. Et. Zucc, Analytical column SOURCE 5RPC ST 4.6/150 was used with a mixture of acetonitrile+water (30+70,volume ratio ) as mobile phase ,with a flow rate of 1ml/min and the UV detection wavelength was 306nm, The linear range was 0.6-1.6mg/ml (r=0.9919). The average recovery of prodatin was 103.0%-90.4%(RSD=7.21%). The method is convenient and accurate. Result indicated a reverse phase chromatography media based o n polystyrene/divinyl benzene has excellent resolution for resveratrol in cuspidatum. P rovide theoretical support for process development.

色谱条件采用SOURCE 5RPC ST 4.6/150预装柱，以乙腈/水(体积比30/70)溶液为流动相，流速1ml/min、检测波长为308nm，结果表明，白藜芦醇在0.6～1.6mg/ml的浓度范围内峰面积积分值与浓度的相关系数r=0.9919；所测定的虎杖饮片中白藜芦醇平均含量为0.304％，平均回收率为103.0％～90.4％；实验表明，以聚苯乙烯为基架的反相介质对于虎杖中自藜芦醇具有良好的分辨率，适合其有效成分的检测、分离，对虎杖中白藜芦醇制备的科学研究过渡到工艺开拓有很好的借鉴。
