筛选法

The DNA was extracted from mature Spinacia oleracea L.separately by the method of CTAB, then used as template after mixed by gender to be amplified with the optimized RAPD reaction condition. A total of 160 primers (10 bp) and 40 pairs of primers were screened, 44 primers of which produced clear patterns.

菠菜为雌雄异株植物，用CTAB法提取其雌雄成株幼嫩叶片总DNA分别构建雌、雄株的DNA池，以之为模板，用已优化的RAPD体系扩增，在160条随机引物(10 bp)和40个引物对中筛选出44个扩增效果较好的单引物，再从中进行进一步筛选。

From the frequently used way that applying probe vectors for choosing promoter to the using of PCR, there were lots of ways for promoter cloning.
