After 24 weeks,the operation area of group A was more smooth,and the surrounding normal cartilage naturally straight flush,transparent form new cartilage,subchondral bone formation in good condition;Group B restoration surgery the basic integrity of the cartilage tissue, center is not yet fully integrated,there was slight depression;CollagenⅡimmunohistochemistry of cartilage that was new brown area.Group C has no formation of articular cartilage.The growth and the intergration of subchondral bone of group A and B were better.

术后24周取材，见A组山羊手术区关节表面较为光滑，与周边正常软骨自然连续平齐，透明的新生软骨组织形成，软骨下骨形成完好；B组山羊手术区修复的软骨组织基本完整，中心部位仍未完全融合，有微小凹陷；Ⅱ型胶原免疫组化示新生软骨组织呈棕黄色。C组术区关节凹陷，无关节软骨组织形成。A组和B组，软骨下骨的生长及与周围组织的结合均较好，无植入物脱落现象的发生。

Depositing cells were incubated in high-sugar DMEM culture medium containing 15% fetal bovine serum, 100 U/mL penicillin, and 100 U/mL Streptomycin, then cells were seeded in a 25 T culture bottle after cell density was adjusted to 1×109 L-1, and incubated at 37 ℃, 0.05 volume fraction CO2 with saturated humidity. After 72 hours, the culture medium was replaced, and red blood cells and other nonadherent cells were removed. Cells began to passage at 70%-80% confluence.

实验方法：穿刺抽取15 mL脐带血，密度梯度法分离吸取分层界面的白色环形絮状物，其中富含单个核细胞，离心后沉积细胞用含15％胎牛血清、100 U/mL青霉素、100 U/mL链霉素的DMEM高糖培养基悬浮，调整细胞密度为1×109 L-1接种于25 T培养瓶内，放置在37 ℃、体积分数为0.05的CO2饱和湿度培养箱中培养，72 h后更换培养基，去除红细胞及其他未贴壁细胞，至70％~80％融合时传代。

RESULTS: During 10 - 12 weeks, in cuntrol group: The defect area was repaired by white and soft tissue that had no resistance to press. The repaired tissue was still lower than the surrounding articular surface with clear boundary. By histological observation, it was found that the defect was repaired by the mechanism similar to inflammatory reaction and the defect is ultimately filled by the hyperplasia of hyaline degenerative fibrous tissues. In filling group: the defect was repaired by semi-transparent, smooth, textured tissues with polish that had resistance to press as well as elasticity. The repaired tissue was almost similar to the shape of the surrounding cartilage,difficult to be distinguished. After histological observation, it was found that there was no inflammatory reaction, but active hyperplasia of inner bonetissue and cartilage tissues; a lot of osteoid tissues and trabeculation were found. Newlborn cartilage was fused with surrounding cartilage tissue and connected with surrounding tissues.

结果：10～12周，对照组：缺损区由白色、质软、按压无阻抗的组织修复，修复组织仍低于周围关节面，边界仍清晰可辨，组织学以类似炎症反应的机制修复缺损，最终以透明变性的纤维组织的增生来填补缺损部位；填充组：缺损区由半透明状、质韧光滑有光泽，按压有阻抗并有弹性的组织修复，修复组织与周围软骨外形上已基本相似，不易区分，组织学未见有炎症反应的过程，内骨组织和软骨组织增生活跃，并可见大量类骨组织和骨小梁形成，新生软骨和周围软骨组织融合，并与周围组织连接。
