To study the applicable prospect of suicide gene on tumor therapy in clinic,we cloned the gene of D-amino acid oxidase and the gene of yeast cytosine deaminase.We hope to establish the transgenic mices of DAAO gene and YCD gene,and the two transgenic strain mices are valuable animal system for studying the biological characteristic of DAAO gene and YCD gene ,for studing the killing activity of DAAO/D-Ala and YCD/5-FC gene therapy systems on tumor.The D-amino acid oxidase gene derived from R.gracilis and it could oxidize D-amino acids. Hydrogen peroxide(H2O2) is a reactive oxygen species generated in the deamination of D-Ala catalyzed by DAAO.

为了探讨自杀基因在临床肿瘤治疗中的应用前景，建立自杀基因肿瘤治疗评价的动物模型，我们克隆了D-氨基酸氧化酶(D-amino acid oxidase，DAAO)基因和酵母菌胞嘧啶脱氨酶(yeast cytosine deaminase，YCD)基因，希望建立DAAO基因及YCD基因转基因小鼠，为研究DAAO及YCD基因的生物学特性、开发和评价DAAO／D-Ala及YCD／5-FC自杀基因系统进行肿瘤治疗建立良好的实验动物模型。

In doing so, we have gotten these objects:(1)built the relationship between natural vegetation pattern of china and the climate,based both on Kira-index and dry/wet index;(2)built the phenology model describing main natural vegetation types of china,based on degree-day;(3)built the drought sensitivity function for main vegetaion types of china; and (4)integrated the drought sensitivity function with the PDSI.

在执行过程中我们基本上按照项目计划书的原初设计完成了如下的工作：①采用Kira的生物热量指数与生物干湿指数相结合的思想，建立了中国自然植被的空间分布与气候因素之间的数量关系；②利用有效积温与生育期之间的关系，建立了自然植被主要类型的物候模型；③根据文献中的实测资料建立了主要农作物类型、自然植被主要类型的缺水敏感性指数；④将此缺水敏感性函数代入Palmer干旱强度指数的计算模型之中。新的干旱强度指数计算步长为周(7天)。

摘要通过分析凝析气藏的相图，建立了凝析液饱和度随压力降的变化规律，并回归得出了相应的公式；同时通过对气体压缩因子、黏度随压力、温度的变化，建立了这些参数和无因次压力的关系；在双重介质地层假设的基础上，建立了在试井过程中凝析气相对渗透率随凝析液饱和度变化的凝析气藏试井解释数学模型，采用隐式迭代的方法进行了求解并进行了参数敏感性分析；结果表明：凝析液的饱和度对凝析气藏试井的压力及压力导数曲线有着很大的影响，早期由于凝析液的析出，阻碍了气藏的流动，导致压力及压力导数曲线上升；在凝析液饱和度达到峰值之后，随着凝析液的挥发，凝析气的相对渗透率逐渐恢复，压力及压力导数曲线又回归到正常的径向流位置；由于裂缝和基岩之间的压力差，使得裂缝弹性储容比在测试过程中发生变化，进而影响了压力及压力导数；窜流系数和裂缝弹性储容比的变化决定了窜流段发生的早晚和程度；而基岩中气体黏度的变化使得窜流的发生稍微滞后。

Based on the previous literatures, the following major innovation works were carried out in this dissertation:(1) The derivatization reaction conditions of ephedrine and pseudoephedrine with sensitive derivation reagent in aqueous system were studied and two new methods were developed for the assay of them by microemulsion electrokinetic chromatography and micellar electrokinetic chromatography with LIF detection. The sensitivity and analysis times were greatly improved compared with the previous reports;(2) A new method of CZE with indirect LIF detection was developed for the simultaneous determination of six coumarin compounds (esculin, esculetin, isofraxidin, genistein, naringin and sophoricoside) with fluorescein as the probe. The proposed method enlarges the application range of LIF detector, and provides new approach for the analysis of certain compounds difficult to derivatize;(3) Based on the above research, an MEKC with indirect LIF detection method for the simultaneous determination of adenine and guanine in DNA extracts from fungus, maize and soybean was established;(4) A new method for the investigation of the complexes formed between human serum albumin and ampicillin sodium under the simulated physiology conditions using laser light scattering technique was developed.

该论文在综述前人工作的基础上，开展了如下未见文献报道的创新性的研究工作：(1)使用灵敏的衍生试剂对麻黄碱和伪麻黄碱在水体系中的衍生反应条件进行了系统研究，建立了微乳电动色谱-激光诱导荧光检测法和胶束电动色谱—激光诱导荧光检测法测定麻黄碱和伪麻黄碱的灵敏分析新方法，与以前的报道相比，灵敏度和分析时间均有很大改善；(2)使用荧光素钠作为背景荧光试剂，建立了同时分析测定六种黄酮类化合物(秦皮甲素、秦皮乙素、异秦皮定、染料木素、柚皮苷和槐角苷)的毛细管区带电泳—间接激光诱导荧光检测新方法，扩大了激光诱导荧光检测的应用范围，对难衍生化合物的分离分析提供了一种新思路和新途径；(3)以荧光素钠作为背景荧光试剂，建立了一种用于同时测定食品提取DNA中的腺嘌呤和鸟嘌呤含量的胶束电动色谱—间接激光诱导荧光检测新方法；(4)用动态和静态激光散射法研究了模拟生理条件下人血清白蛋白与氨比西林钠盐相互作用所形成的复合物，为药物分子与蛋白质的相互作用、药代动力学等的研究提供了一种新思路。

We mainly concentrate on the role of Chinese etymologic relations of lexemes played in setting up inter-lexeme definitions, and on the role of both English and Chinese derivatives played in inter-lexeme definitions.

Firstly, the author establish the functions of stability on the basis of the spline function method ,according to the theory of geometrical nonlinear and the theory of stability, educe the stability equation of structure based on variational principle, then carry through the flexuosity analysis of CFST arch.The big distortion geometrical nonlinear was considered by the stability equation,the author simplified the problem of geometrical nonlinear through ignored quadratic term and reserve the simple term, at last boiled down to solve the linear stability equation of eigenvalue .

利用样条函数方法、根据几何非线性变形理论和结构稳定性理论，先建立起结构稳定的泛函，再由变分原理导出结构的稳定方程，进行钢管混凝土拱屈曲分析；本文所建立的稳定方程考虑了几何非线性的大变形效应，通过忽略几何非线性的二次项，保留一次项，将几何非线性问题简化，最后归结为求解线性稳定问题的特征值稳定方程，建立了结构非线性静力稳定性问题的新算法及失稳类型判别准则。

By combining the principal component analysis and cluster analysis techniques, we select five factors and group design storm hyetograph into three categories in the study area. Finally, we employ the method of analytic hierarchy process to establish the regionalized representative hyetograph for Northern Taiwan area.

本研究针对台湾北部地区提出一个新的方法来评估并建立区域代表雨型，首先建立各测站具尺度不变性之高斯马尔柯夫雨型，其次利用主成份分析法萃取出雨型中的5个重要的控制因素，再就这些因素以聚类分析法将研究区域内之设计暴雨雨型分为3个均匀群集，但同一群集内之雨型并非唯一，故研究中最后利用层级分析法进行评估，选取并建立出区域代表雨型。

ABSTRACT Aiming at problems of trans-vector controller in a vehicle-carried induction motor such as variable rotor and stator resistance following motor temperature, and their inductance varying with flux saturation degrees. Firstly, CSI and VSI are introduced, for CSI, it can hold the advantages both FOC and DTC using stator flux directional, PI parameter and simulation are also introduced. Secondly, equations of IM in the synchronous rotary frame and its static counterpart, as figuring in the saturation condition of magnetic field, are dealt in this paper and the simulation results are presented. Thereafter, the parameter designing of PI trans-vector controller is processed, which features the non-linear model. In practical realization, as neglecting the dynamical change of motor inductance, this paper employs online modification of inductance parameters, according to the flux saturation, then transforms the non-linear equations into linear ones, and so compacts the structure of controller. Thirdly, in view of the maximum torque output is extraordinary requisite when Electrical Vehicle start up, accelerate, and overtake, this paper issues the maximum torque when both inverter and battery capacity is corporeally definite. Fourthly, on-line differentiating and analyzing both rotor's time constants and its flux by using the expanding order reduction and discrete Kalman filter equation are fathomed, in order to realize high-performance trans-vector algorithm, and also the simulation results are presented. Fifthly, the close-loop regenerative brake system, when the EV's torque is set constant, is deduced; at the same time, unification of electric drive model and brake model is executed. Combined with maximum charge current demands, the mathematical model for online yielding torque as a demand.

本论文针对车载异步电机在矢量控制器所遇到的问题：定转子电阻随温度变化及随电机饱和程度变化的转子电感及定子电感而影响电机调速性能提出一套完整的解决方案：分析了电流型逆变器和电压型逆变器在实现矢量控制时控制器参数的计算，分析出对于电流型逆变器在采用定子磁场定向时，其性能同时具有矢量控制和直接转矩控制两者的优势，并对电压型逆变器在转子磁场定向下的模型进行了仿真研究；建立异步电机非线性模型，推导出考虑磁饱和时异步电机在同步旋转坐标系及静止坐标系下的方程，并做出了仿真结果，建立异步电机在非线性模型下的矢量控制调节器的PI参数设计，在实际应用中，若忽略电机电感的动态变化，可以根据当前的磁路饱和状态而在线修正电感参数，从而将非线性方程线性化，降低控制器的复杂度；考虑到电动汽车在起动和加速超车时需要电机有最大转矩输出，本文讨论在逆变器容量一定和电池供电能力有限的情况下电机最大转矩输出问题；推导了异步电机扩展降阶、离散卡尔曼滤波方程在线辨识转子时间常数和转子磁通，用于实现高性能的矢量控制算法，并给出了仿真结果；推导出了电动汽车恒转矩给定的闭环回馈制动系统，实现了电动控制模型和制动模型的统一，而且结合铅酸电池最大充电电流的要求，为制动转矩在线给定建立了数学模型；设计了基于双DSP系统的高性能矢量控制器软硬件框图，并以大量实验数据说明矢量控制在电动汽车应用的实际应用状况。
