To identify whether our previous founding were exist in the series of our newly established cell cultures with the same genetic background and different metastatic potentials, and to make clear what are congenerous and various traits in the chromosomal or DMA sequence levels between these cells, in this study, a combination of conventional G-banding, comparative genomic hybridization, multiplex fluorescence in situ hybridization and arm or locus-specific fluorescence in situ hybridization were used to comprehensively characterize molecular cytogenetics aberrations of the above cell cultures.

为查明我所新建立的这些具有同一遗传背景而又在肝癌的转移潜能上存在明显差异的细胞是否也具有8p的缺失，并弄清它们在其它染色体及其区段的DNA水平上有着怎样的相似和不同，这些相似与不同又有着怎样的意义，本课题联合应用G显带技术、比较基因组杂交技术、多重荧光原位杂交技术和位点或臂特异荧光原位杂交技术，对上述细胞进行细胞及分子遗传学研究，以期了解其分子细胞遗传学异常的特征、找出转移相关的遗传学标记，为探讨肝癌转移机制提供材料。

Closer to the official opening ceremony, people were starting to gather in Plaza Consistorial, where the ceremony was held, and the adjoining Plaza Castillo. Plaza Consistorial was transformed into a big, open-air party. People were chanting, tossing large balloons and splashing each other with red wine or champagne. Those on the balconies around the square also joined the folly and treated those below them with buckets of water, one after one. It all sounded fun until the crowd became too close for comfort and some people had the clever idea of pushing the others in all directions, while ground was becoming ever more slippery. The whole scene was indeed on a slippery slope itself!

Base on core observation and grain size analysis, and under associated consideration of logging trace characteristics, Chang8 reservoir in study area is classified into three sedimentatal microfacies, subsea river, mouth bar and river flank.3.The mian diagenesis in study area are: mechanical compaction, cementation, metasomasis and denudation in late diagenetic phase.4. In allocation of reservoir macroscopic heterogeneity, more heterogeneticer reservoir, class 1 and class 2, mainly locate in the center of subsea river or crook and divaricate of the river; class 3 and class 4 mainly locate in river flank, and the physical property of these locality are better.5. Diagenesis of reservoir have influence on the pore structure which influences the reservoir microscopic heterogeneity. The better the pore structure is, the slighter the reservoir microscopic heterogeneity is; on the contrary, the reservoir microscopic heterogeneity is serious.

本次研究所取得的主要成果及熟悉有：1、将合水地区长8储层细分为6个小层：长8_1~1、长8_1~2、长8_1~3、长8_2~1、长8_2~2和长8_2~3.2、依据岩心观察和粒度分析，并结合测井曲线特征，研究区长8储层分为水下分流河道、河口坝和河道侧翼四种沉积微相。3、研究区主要经历的成岩作用有：机械压实和压溶作用、胶结作用、交代作用、晚成岩期溶解作用。4、从宏观非均质性平面展布特征来看研究区宏观非均质较强的1类和2类区主要分布于水下分流河道中间或河道拐弯和分又处。3类和4类往则主要分布于水下分流河道侧翼，这些地方储层均质性相对较好。5、储层成岩作用影响储层的孔隙结构，储层的孔隙结构又对储层的微观非均质性有不同程度的影响，孔隙结构越好，则储层的均质性越好，储层的孔隙结构差，则非均质性强。

Chronicity maybe result from many factors with complex mechanism.HCV may evade the host immune surveillance due to the hypermutation of HCV gene; HCV has the nature of pantropic,the tissues outside liver can provide virus for repeatedly infection of liver; The HCV receptors,such as CD81,LDL receptor,scavenger receptor B type I,L-SIGN,DC-SIGN and so on,may influence organism humoral immunity and the cellular immunity function in different degrees; HCV infection also may cause the reduction of quantity of organism immune cell or functional defect,low response of humoral immune response,tolerance of cellular immunity,then,the host finally transforms into chronic infection condition.

Egr-1 mRNA and Egr-1 protein hadn't been found in the normal vein. The expressions of Egr-1 mRNA and Egr-1 protein had biphasic changes. By reverse transcription-PCR and in situ hybridization, we found that the level of Egr-1 mRNA rose at 1 hour after graft, the expression of Egr-1 mRNA was (35±7)%. Decline at hour 6, 24 and day 3, the positive rates of Egr-1 mRNA were (8±2)%,(8±6)% and (8±4)% respectively. Reincrease at day 7, a peak at day 28, the positive rate of Egr-1 mRNA was (45±6)%(compared with other phase, P＜0.01). At day 42, the expression of Egr-1 mRNA declined again. Immunohistochemical staining and Western blot revealed Egr-1 protein had expressed at hour 2 early phase, the expression of Egr-1 protein was (30±5)%, and until to hour 6. The level of Egr-1 protein was decrease at hour 24 and day 3, the positive rates were (7±3)% and (7±8)% respectively.

结果 自体静脉移植后，Egr-1 mRNA和Egr-1蛋白的表达呈双相变化，即移植后1 h， Egr-1 mRNA表达迅速升高，阳性率为（35±7）%，6 h、24 h及3 d时下降到较低水平，阳性率分别为（8±2）%、（8±6）%和（8±4）％，7 d时又再升高，28 d时达高峰，阳性率为（45±6）%，此与其余各时点比较差异均有统计学意义（P＜0.01），42 d时，Egr-1 mRNA的表达再次下降；移植早期（2 h）即有Egr-1蛋白的表达，阳性率为（30±5）%，并持续至6 h，24 h～3 d表达下降到较低水平，阳性率分别为（7±3）%和（7±8）%，7 d时又再升高，至移植后28 d，Egr-1蛋白的表达阳性率达到高峰，为（40±9）%，此与其余各时点比较差异有统计学意义（P＜0.01）。
