Firstly, the establishment of model requires sophisticated neurointerventional techniques and costly interventional materials(sacculus, minikin circlip ring, endovascular electrocoagulation), it not only consumed time but also increased the resesrch expense ; Secondly, although the Massoud model is the first successful model that contain the nidus, but it is far from mimicking the high-flow fistulae frequently observed in a brain AVM; Thirdly, the rapid spontaneous thrombosis result in fistula occlusion in a short time;Fouthly, the model's natural course and the long–term patency of fistula are unclear;Currently the model is only used for the short-term research.

但Massoud模型目前存在以下主要缺点：第一，模型的建立需要复杂的神经介入技术和昂贵的介入材料(球囊、微弹簧圈、血管内电凝等)，操作费时，又大大增加了研究费用；第二，Massoud模型虽然首次具备了畸形血管团，但它远不是我们临床上常见的含有高流量动静脉瘘的脑AVM；第三，颈动静脉吻合口自发性血栓形成进而导致引流静脉闭塞的机率很高；第四，Massoud模型的长期自然过程和动静脉吻合口的长期通畅性尚不清楚，目前仅仅作为急性期模型。

Also this study documents the overexpression of MMP3 which may degrade ECM in the pathogenesis of the pterygium.

结论翼状胬肉组织中ECM的成分较正常球结膜发生改变，有大量的弹性纤维变性、胶原纤维增生及新生血管形成；col Ⅲ在翼状胬肉组织中表达增加，LN及胶原纤维可能对翼状胬肉中血管形成、细胞粘附和迁移及ECM重建起作用；MMP-3在翼状胬肉组织中表达增加，其对ECM成分的降解作用可能有助于翼状胬肉的形成和向角膜中央侵袭。

Thirty patients with acute promyelocytic leukemia were observed between September 1977 and October 1985. Bleeding diathesis with petechiae or ecchymosis (80%) was the most common presenting symptom. Seventeen of 24 patients with pretreated coagulation studies were diagnosed to have disseminated intravascular coagulation. Six out of 11 patients died from intracranial hemorrhage after a mean of 2.5 days of hospitalization without any treatment. DIC was responsible for death in most cases. Induction therapy with anthracycline and cytosine arabinoside combined with prophylactic heparin infusion was carried out in the remaining 19 patients. Two patients were incompletely treated. Eight out of 17 patients achieved complete remission after a mean of 63 days following chemotherapy. The remission rate was 47%.

自1977年9月至1985年10月，观察30位急性前骨髓球性白血病病帣，临床上诊断时初发症状，主要为出血现象，尤其以瘀斑出血最多，占80%。24位病人作治疗前凝血机能检查，17位出现播散性血管内凝固症候群。11位病人没有接受治疗，其中6位经诊断后平均2.5天内死於脑内出血，凝血机能检验显示5位有播散性血管内凝固症候群。19位病人接受anthracycline与ara-C并用预防性肝素静脉滴注作缓解诱导治疗，2位不完全治疗，其余17位中有8位经过平均63天之化学治疗得到完全缓解，缓解率47%。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.

研究方法：(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC，细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构；(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定；(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC，并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况，根据各个细胞克隆受Dox调控表达的程度，选择低背景、高诱导表达AT2R的细胞系，作为双重稳定MSC细胞系；蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况；(4)建立大鼠颈动脉球囊损伤动物模型，将双重稳定MSC在术中导入血管，分别于14 d、28 d进行病理切片，检测可调控AT2R对新生内膜增生的影响；采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变，TUNEL法检测血管组织中细胞凋亡的变化情况。

Results There was a similar distributive pattern of Neul, PPCA and β-gal in the inner ear. Neul intense staining was observed in the cochlear spiral ganglion cells, spiral limbus, spiral ligament, vestibular ganglion cells, cristae, maculae hair cells, and weak staining in inner hair cells, outer hair cells, supplying cells of the organ of Corti and stria vascularis. The intense staining of PPCA and β-gal were observed in the spiral ganglion and vestibular ganglion cells, and weak staining in the spiral limbus, spiral ligament, stria vascularis and organ of Corti. The inner ear exhibited no staining when Neul, PPCA and β-gal were deficient, respectively.

Neul最强的染色主要在螺旋神经节细胞、螺旋韧带、螺旋缘、前庭神经节细胞及壶腹嵴、球囊和椭园囊感觉毛细胞，较弱的染色分布于血管纹和Corti器内、外毛细胞及支持细胞；PPCA和β-gal在螺旋神经节和前庭神经节细胞有较强的染色，血管纹、螺旋韧带、螺旋缘和Corti器内、外毛细胞及支持细胞呈较弱的染色反应；各自酶缺乏时内耳免疫染色消失。
