To acquire more information about gene expression at the whole-genome scale,our lab and National Engineering Center for Beijing Biochip Technology worked together and constructed the first genome-wide oligonucleotide microarray of the silkworm.This microarray was used to detect gene expression of 10 tissues/organs,including midgut,integument,head,hemocyte,testis,ovary,anterior/median silk gland,posterior silk gland,fat body and malpighian tubule.In this study,we performed analysis of these microarray data and confirmed the reliability of the results.

RESULTS: FGFR1-positive chondrocytes were abundant in the maturative layer and hypertrophic layer, but only few FGFR1-positive chondrocytes were found in the proliferative layer. In the control group, the expression of FGFR1 increased from 6-week-old to 10-week-old rats, and then decreased and went stable. In both young and adult group, the expression of FGFR1 in the experimental group was significantly lower at 4, 6 weeks and higher at 8 weeks than that in the control group, especially in the young group (P.05). But no difference was found between the experimental group and the control group at 2 weeks in both groups. CONCLUSION: FGFR1 may play an important role in the remodling of condylar cartilage induced by occlusal disorders.

结果：FGFR1主要表达在大鼠髁突软骨过渡层和肥大层，增殖层很少；正常组大鼠髁突软骨从6周龄到10周龄FGFR1表达逐渐增强，10周龄后降低，并保持在一个稳定水平；成年组和幼年组实验4、6周时FGFR1的表达均显著低于对照组，实验8周时的表达高于对照组(P.05)，幼年组尤为明显，实验2周组与对照组无显著差异。

Our previous studies have shown that ubiquitous overexpression of HmgD caused hemocytes proliferation and the formation of melanotic tumors in the Drosophila larvae. To clarify the mechanism of melanotic tumors appearance,we also generated RT-PCR analysis to test the transcriptional changes of PPAE and Spn27A,which encode the key factors in the phenoloxidasecascade pathway from larvae samples of mid-third instar,late-third instar and prior to pupation relative to wild type flies.Our result showed that in the HmgD overexpression mid-third instar larvae,the expression level.of Spn27A was lower than wild type,otherwise the expression level of PPAE was higher than wild type.It suggests that overexpression of HmgD affects the PO cascade pathway.

本研究组前期工作发现，过量表达HmgD引起果蝇幼虫体内血细胞增生，并出现黑色素瘤，为探讨HmgD过量表达导致了血细胞增生即形成黑色素瘤的分子机制，本研究分别提取三龄中期、后期和相对于野生型化蛹前时刻的幼虫的RNA，首先检测了编码酚氧化酶级联反应途径中的两个关键基因PPAE，Spn27A在突变体中的转录发生变化，结果发现，Spn27A在三龄中期的表达量低于野生型，而PPAE的表达高于野生型，表明过量表达HmgD影响了酚氧化酶级联反应途径，低水平的Spn27A不足以抑制PPAE的活性，活化的PPAE催化了PPO的活性，使其转变成有活性的PO，从而促进黑色素的生成，使果蝇体内产生黑色素瘤。

Results: The expression of HO-1 mRNA in macrophage cell was significantly enhanced by the melilotus suaveolens extract. The result of Western blot showed that the expression of HO-1 protein was significantly increased after the intervention of melilotus suaveolens extract.
